مطالعه ارزیابی هورمون تستوسترون و شاخص‌های اسپرماتوژنز در موش‌های صحرایی دیابتی تحت تیمار عصاره آبی زعفران (Crocus sativus L.)

مطالعات پیشین نشان می‌دهد که، ترکیبات گیاهی می‌توانند در درمان بسیاری از بیماری‌ها از جمله دیابت موثر باشند. دیابت یک بیماری متابولیکی جدی است که می‌تواند باعث اختلال در عملکرد طبیعی سیستم تناسلی شود. به همین منظور، تاثیر عصاره آبی زعفران بر سطح هورمون تستوسترون و شاخص‌های اسپرماتوژنز در موش‌های صحرایی دیابتی شده نژاد ویستار مورد بررسی قرار گرفت. در این مطالعه ۳۵ سر موش صحرایی نر نژاد ویستار به ۵ گروه ۷ تایی تقسیم شدند. جهت القای دیابت از داروی استرپتوزوتوسین به میزان ۹۰ میلی‌گرم بر کیلوگرم استفاده شد. جهت ارزیابی اثر عصاره زعفران، درمان حیوانات به مدت ۳۵ روز ادامه پیدا کرد. متعاقب پایان یافتن دوره درمان، حیوانات توسط بیهوشی عمیق آسان کشی شدند. نمونه خون جهت اندازه‌گیری سطح سرمی تستوسترون، جمع‌آوری شد. سپس، نمونه برداری از بافت بیضه انجام شد و مراحل روتین آماده‌سازی مقاطع بافتی انجام پذیرفت. در نهایت نمونه‌ها، جهت ارزیابی‌های هیستومورفومتریک بیضه آماده شدند. علاوه بر اینها، شاخص تمایز لوله‌ای و شاخص اسپرمیوژنز محاسبه گردید. همچنین تحرک و زنده مانی اسپرم، بررسی شد. نتایج نشان داد که در گروه دیابتی ضخامت اپیتلیوم و قطر لوله‌های اسپرم ساز کاهش یافته بود و عصاره زعفران با دوزهای ۱۲۰ و ۲۴۰ میلی‌گرم سبب بهبود آسیب حاصله شده بود. دیابت سبب کاهش سطح تستوسترون سرمی شده بود که عصاره زعفران خصوصا با دوز ۱۲۰ میلی‌گرم سبب افزایش قابل توجه این هورمون شده بود. مشاهدات نشان داد که، عصاره زعفران بدلیل خاصیت آنتی اکسیدانی می‌تواند باعث کاهش اثرات منفی دیابت بر روی بافت بیضه و خصوصیات هیستومورفومتریک آن شود.

Previous studies show that herbal compounds can be effective in the treatment of many diseases, including diabetes. Diabetes is a serious metabolic disease that can disrupt the normal function of the reproductive system. For this purpose, the effect of aqueous saffron extract on the level of testosterone hormone and spermatogenesis indices in diabetic Wistar rats was investigated. In this study, 35 male Wistar rats were divided into 5 groups of 7 each. In order to induce diabetes, the drug streptozotocin was used in the amount of 90 mg/kg. In order to evaluate the effect of saffron extract, the treatment of animals continued for 35 days. After the end of the treatment period, the animals were euthanized by deep anesthesia. Blood samples were collected to measure the testosterone serum level. Then, sampling of the testicular tissue was done, and the routine steps of tissue section preparation were performed. Finally, the samples were prepared for the histomorphometric evaluations of the testis. In addition to these, Tubular Differentiation Index (TDI) and Spermiogenesis Index (SI) were calculated. Also, sperm motility and viability were analyzed. The results showed that in the diabetic group, the thickness of the epithelium and the diameter of the spermatogenic tubes decreased, and the saffron extract with doses of 120 and 240 mg improved the damage. Diabetes caused a decrease in serum testosterone level, and saffron extract, especially at a dose of 120 mg, caused a significant increase in this hormone. Observations showed that saffron extract can reduce the negative effects of diabetes on testicular tissue and its histomorphometric characteristics due to its antioxidant properties.
