بررسی تنوع ژنتیكی برخی گونه های زعفران ایران با استفاده ازخصوصیات کروموزومی

زعفران وحشی از گونه های جنس Crocus ، گیاهانی چند ساله، علفی و زینتی از خانواده زنبقیان Iridaceae هستند. مجموعهCrocus cancellatus به عنوان گونه وحشی زعفران دارای تنوع زیادی در مورفولوژی، سیتولوژی و اکولوژی می باشد. این پژوهشبه جهت بررسی خصوصیات کروموزومی و تهیه کاریوتیپ چهار اکوتیپ زیر گونه damascenus گیاه زعفران وحشی انجام شدهاست. تکامل کاریوتیپ میتواند از طریق تعیین خصوصیت مورفولوژی کروموزوم بر اساس موقعیت سانترومر بازسازی شود. در اینتحقیق بنه های چندین جمعیت از Crocus cancellatus subsp. Damascenus از استان های کردستان، مرکزی، فارس و لرستانجمع آوری شده و سپس در گلخانه دانشگاه کردستان کشت شدند و از ریشه های در حال رشد برای مطالعات سیتوژنتیکی استفاده گردید.بهترین تکنیک برای مطالعه میتوز، استفاده از بافت های مریستم نوک ریشه برای آمادهسازی کاریوتایپ می باشد. پارامترهای کروموزومیشامل طول بازوی بلند (L)، طول بازو کوتاه (S)، طول کروموزوم (CL)، نسبت بازوها (L/S, AR) و شاخص سانترومری (CI = S / CL) تخمین گردید. روش همبستگی برای آنالیز کلاستر برای بررسی شباهت بین جمعیت ها از نظر صفات مورفولوژی و کروموزومیاستفاده شد. بررسی های انجام شده نشان داد هر کدام از جمعیت ها دارای تعداد کروموزوم متفاوت می باشند. به طوریکه جمعیت اقلیدn=21 و جمعیت الیگودرز n=8 و مریوان n=8 و سقز دارای n=21 کروموزوم میباشند. نتایج حاصل از بررسی تنوع ژنتیکی وهمچنین بررسی فواصل جغرافیایی و دوری و نزدیکی ژنوتیپ های مورد مطالعه نشان داد که به طور کلی فواصل جغرافیایی دلیلی بردوری و نزدیکی ژنتیکی افراد نمی باشد و نتایج بیانگر ژنوتیپ های مربوط به استان کردستان (مریوان و سقز) درون یک گروه واقع نشدهو در دو گروه مجزا قرار گرفته اند.

Wild saffron, from the genus Crocus, consists of perennial, herbaceous, and ornamental plants belonging to the Iridaceae family. The Crocus cancellatus complex, as a wild saffron species, exhibit  great diversity in morphology, cytology, and ecology. This study was conducted to investigate chromosome characteristics and to prepare a karyotype of four ecotypes of the subspecies damascenus of wild saffron. Karyotype evolution can be reconstructed by determining the morphological characteristics of chromosomes based on centromere position. In this research, corms from several populations of Crocus cancellatus subsp. damascenus were collected from the provinces of Kurdistan, Markazi, Fars, and Lorestan, then cultivated in the greenhouse of Kurdistan University, and roots in active growth were used for cytogenetic studies. The best technique for studying mitosis is using root tip meristem tissues for karyotype preparation. Chromosomal parameters including long arm length (L), short arm length (S), chromosome length (CL), arm ratio (L/S, AR), and centromeric index (CI = S / CL) were estimated.The correlation method was used for cluster analysis to examine the similarity between populations in terms of morphological and chromosomal traits. The investigations showed that each of the populations has a different number of chromosomes. Specifically, the Eqlid population has n=21, the Oligodorz population n=8, Marivan n=8, and Saqez has n=21 chromosomes. The results of genetic diversity analysis, as well as the examination of geographical distances and the proximity or remoteness of the studied genotypes, indicated that, in general, geographical distances are not a reason for the genetic closeness or distance among individuals. The results also showed that the genotypes related to Kurdistan province (Marivan and Saqez) did not fall within the same group and were instead placed in two separate groups.
