زعفران، که از کلاله‌های خشک‌شده گیاه Crocus sativus L. گرفته می‌شود، به دلیل رنگ، عطر و خواص دارویی منحصر به فردش به عنوان “طلای سرخ” شناخته می‌شود و آن را به یکی از ارزشمندترین ادویه‌های جهان تبدیل کرده است. این ارزش بالا در درجه اول به آپوکاروتنوئیدهای چند منظوره آن، به ویژه کروستین، کروسین، پیکروکروسین و سافرانال نسبت داده می‌شود که در ویژگی‌های حسی و فعالیت‌های زیستی آن نقش دارند. به طور سنتی، این ترکیبات مستقیماً از گیاه استخراج می‌شوند، فرآیندی که هم پرزحمت و هم با بازده کم است. علاوه بر این، C. sativus در مناطق خشکی مانند ایران، کشمیر (هند) و اسپانیا رشد می‌کند، اما کشت آن نیاز به آبیاری قابل توجهی دارد. این نیاز می‌تواند باعث تشدید کاهش آب‌های زیرزمینی و تشدید رقابت برای منابع آب در این مناطق شود. در پاسخ به این چالش‌ها، پیشرفت‌ها در مهندسی متابولیک و زیست‌شناسی مصنوعی، بازسازی مسیرهای بیوسنتزی آپوکاروتنوئید زعفران را در میزبان‌های ناهمگن امکان‌پذیر کرده و تولید پایدارتر و مقیاس‌پذیرتر را ممکن ساخته است. این فصل مروری بر آپوکاروتنوئیدهای اصلی زعفران ارائه می‌دهد و پیشرفت‌های حاصل در تولید آنها را با استفاده از کارخانه‌های میکروبی و سلولی گیاهی برجسته می‌کند. علاوه بر این، محدودیت‌ها و چالش‌های فعلی مرتبط با بیوسنتز آپوکاروتنوئید زعفران مبتنی بر کارخانه سلولی نیز مورد بحث قرار می‌گیرد.

Saffron, derived from the dried stigmas of Crocus sativus L., is esteemed as “red gold” due to its unique color, aroma, and pharmacological properties, making it one of the world’s most valuable spices. This high value is primarily attributed to its multifunctional apocarotenoids, notably crocetin, crocin, picrocrocin, and safranal, which contribute to its sensory characteristics and bioactivities. Traditionally, these compounds are extracted directly from the plant, a process that is both labor-intensive and with low yields. Moreover, C. sativus thrives in arid regions such as Iran, Kashmir (India), and Spain, yet its cultivation demands substantial irrigation. This requirement can exacerbate groundwater depletion and intensify competition for water resources in these areas. In response to these challenges, advancements in metabolic engineering and synthetic biology have enabled the reconstruction of saffron apocarotenoid biosynthetic pathways in heterologous hosts, enabling more sustainable and scalable production. This chapter provides an overview of the major saffron apocarotenoids and highlights breakthroughs in their production using both microbial and plant cell factories. Additionally, the current limitations and challenges associated with cell factory-based saffron apocarotenoid biosynthesis are also discussed.
